Synthesis of aflatoxins by the non-growing mycelia of Aspergillus parasiticus and the effect of inhibitors.

چکیده
A chemically defined medium giving high yields of aflatoxins has been reported from this laboratory (Reddy, Viswanathan & Venkitasubramanian, 1971). Many workers have studied the incorporation of [ 1J4C]acetate into aflatoxins (Adye & Mateles, I 964; Donkersloot, Hsieh & Mateles, 1968; Detroy & Hesseltine, 1969; Hsieh & Mateles, 1970, 1971). Labelled acetate is efficiently incorporated into aflatoxins when added directly to cultures of Aspergillus parasiticus. When studying the effects of different factors on aflatoxin production with such a system, it is always possible that the results will be influenced by the growth of the mycelium. Therefore, attempts were made to develop a system capable of synthesizing aflatoxins under completely resting conditions, and this system was used to study the effect of different inhibitors on aflatoxin production.

Aflatoxin biosynthesis in Aspergillus parasiticus: effect of methionine analogs.
The physiological effects of DL-ethionine and related methionine analogs upon cellular protein synthesis and methyl group transfer to aflatoxin B1 in Aspergillus parasiticus were examined in growing and resting cell systems. The addition of DL-ethionine (0.02 M) to growing mycelia inhibits cellular growth some 37% after 120 h of development. Addition of ethionine before 50 h prevents any aflato...
متن کاملMolecular identification of aflatoxigenic Aspergillus species in feedstuff samples
Background and Purpose: Aflatoxins are naturally produced by some species of Aspergillus, such as A. flavus and A. parasiticus. Aflatoxins reportedly have carcinogenic effects on human, poultry, and livestock, and therefore could be linked to severe human illnesses. Aflatoxin biosynthesis pathway involves different clustered genes, including structural, regular, and unassigned genes. The presen...
متن کاملInhibitory Effects of Silver Nanoparticles on Growth and Aflatoxin B1 Production by Aspergillus Parasiticus
Background: Aflatoxins (AFs) are secondary hazardous fungal metabolites that are produced by strains of some Aspergillus species on food and feedstuffs. Aflatoxin B1 (AFB1) is one of the most important AF with high toxicity. Prevention of AF production and their elimination from food products is a matter of importance for many researchers in the last decades.Nanomaterials applications in medica...
متن کاملComparative Study of the Major Iranian Cereal Cultivars and some Selected Spices in relation to Support Aspergillus parasiticus Growth and Aflatoxin Production
Background: Aflatoxins are toxic fungal metabolites enable to contaminate a wide range of natural substrates. This contamination can be host-specific for different plant species. In this study, the ability of a toxigenic Aspergillus parasiticus to produce various aflatoxins on major Iranian cereals was evaluated with special focus on plant susceptibility to toxin production at cultivar level. M...
متن کاملCharacterization of aflatoxigenic Aspergillus flavus and A. parasiticus strain isolates from animal feedstuffs in northeastern Iran
Aflatoxins are secondary toxic metabolites produced by some Aspergillus spp. particularly, Aspergillus flavus and A. parasiticus that contaminate food and feed. The objective of this study was to evaluate the contamination of feedstuffs with Aspergillus spp. and detect genes involved in the aflatoxin biosynthesis pathway of A. flavus and A. parasiticus isolates. A total of 110 cow feed samples ...
